Propidium Iodide (PI) is a red fluorescent, cell-impermeable nucleic acid stain widely used to identify dead cells in mixed live-dead populations. Its fluorescence increases 20–30× upon binding to nucleic acids, making it highly sensitive for detecting DNA in mammalian cells, bacteria, and yeast. PI is compatible with flow cytometry and fluorescence microscopy and is often employed as a counterstain in multicolor experiments. Key spectral properties: λAbs/λEm 535/617 nm (DNA-bound) and 493/636 nm (unbound).
